Police Chief Fred Westerbeke said that the shooter wore a bulletproof vest as he shot and killed the female victim and wounded her teenage daughter.
The first shooting took place at a house on the west side of the city on Heiman Dullaertplein – close to the home of the suspect.
The shooter then went to the nearby Erasmus Medical Center where he shot and killed a teacher in a classroom, Westerbeke said.
Two major fires were also started at both locations.

Cops are still investigating the suspect's motive behind the twin shootings.
He was known to police after being convicted of animal abuse in 2021 public prosecutor, Hugo Hillenaar, said in tonight's press conference.
The attacks sparked mass panic as hospital staff, students and patients were forced to run for their lives to escape from the gunman and the blaze.
Footage from the scene showed heavily armed police units wearing body armour as they stormed the hospital, while helicopters buzzed overhead.
Cops say the situation at Erasmus Medical Center is now under control.
The suspect, was described in local media reports as a "tall man, dressed in combat clothing carrying a rucksack and wearing headphones".
Police say he used a handgun and reportedly fled the scene on a motorcycle.
"First there was a shooting on the fourth floor. Four or five shots were fired. Then a Molotov cocktail was thrown into the education centre," a medical student told RTL Nieuws.
The only suspect is now in police custody after an urgent manhunt was launched to locate him.
The man appears to have been apprehended at the city centre hospital's helipad.
